摘要:
心肌细胞凋亡在心肌缺血再灌注损伤、心肌梗死后心室重塑、心肌病和心力衰竭等病理生理过程中具有重要作用。哺乳动物不育系20-样激酶1(Mst1)是一种丝氨酸/苏氨酸蛋白激酶,属于生发中心激酶亚家族Ⅱ中的一员。作为Hippo信号通路上游的关键分子,Mst1具有调控细胞增殖和凋亡,影响组织器官生长等作用。在病理情况下,Mst1具有促进心肌细胞凋亡的作用。心肌细胞特异性Mst1过表达小鼠具有扩张型心肌病的表型;敲除或抑制内源性Mst1则可抑制心肌细胞凋亡,改善心功能。现主要综述了Mst1促进心肌细胞凋亡的作用,以及β肾上腺素能受体通过Mst1调控心肌细胞凋亡的分子机制,并评估了Mst1敲除或失活对心脏可能产生的保护作用。
Abstract:
Cardiomyocyte apoptosis plays an important role in the pathophysiological process of several cardiac disorders,such as myocardial ischemia/reperfusion injury,myocardial infarction and remodeling,cardiomyopathy as well as heart failure. Mammalian sterile 20-like kinase 1 (Mst1) is a serine-threonine kinase and belongs to the germinal center kinase subfamily Ⅱ. As an upstream component of Hippo signaling pathway,Mst1 plays an important role in regulating cell apoptosis and proliferation. Recent studies found that Mst1 could induce cardiomyocyte apoptosis under pathological conditions. Transgenic mice with cardiomyocyte specific overexpression of Mst1 have dilated cardiomyopathy phenotype including chamber dilatation,systolic dysfunction and fibrosis. Mst1 deletion or inactivation can inhibit cardiomyocyte apoptosis and improve cardiac function. In this review,we summarized the promoting role of Mst1 in cardiomyocyte apoptosis,and updated the current knowledge of cardiac β-adrenoceptors/Mst1 signaling pathway on cardiomyocyte apoptosis. The protective effect of Mst1 knockout or inactivation on the heart was also evaluated in this review
