摘要:
抗凝药物在治疗和预防血栓栓塞性疾病中发挥着关键作用,通过干扰凝血级联反应的关键环节,有效控制血栓形成。然而,传统抗凝药物在减少血栓的同时,常伴有不同程度出血风险的增加。以凝血因子Ⅺ为靶点开发的新型抗凝血药—Ⅺ因子抑制剂可通过抑制Ⅺ因子来阻断凝血酶的生成,进而抑制血栓形成,且对生理性止血的影响较小,已成为最有潜力的新型抗凝血药之一。现对以Ⅺ因子为靶点的抗凝药物的病理生理机制及最新临床研究进展进行综述,以期为血栓栓塞性疾病的防治提供更多的选择。
Abstract:
Anticoagulant drugs play a crucial role in the treatment and prevention of thromboembolic diseases,effectively controlling thrombosis by interfering with key steps in the coagulation cascade. However,traditional anticoagulant drugs are often associated with an increased risk of bleeding to varying degrees while reducing thrombosis. Factor Ⅺ inhibitors,a new class of anticoagulants,target coagulation factor Ⅺ and block thrombin generation and inhibit thrombosis while having less impact on physiological hemostasis,which makes them as one of the most promising new anticoagulants. This article reviews the pathophysiological mechanisms and the latest clinical research progress of factor Ⅺ inhibitors,aiming to offer safer and more effective treatment options for patients with thromboembolic diseases.
